Elabscience®化学发光 (ECL) 底物液是一款辣根过氧化物酶 (HRP) 底物,可用于检测辣根过氧化物酶(HRP)标记的蛋白或核酸,用于蛋白质印迹或ELISA等分析蛋白含量,可兼容胶片和数字显影系统。

使用说明
1. WB检测
1) 按照免疫印记实验要求封闭印记膜,孵育一抗和相应的二抗。
2) 用 TBST 洗膜3次,每次 5 min。
3) 根据需要吸取适量底物A液和底物B液1:1等量混合。
注:该试剂现配现用,A液和B液在吸取过程中必须要更换吸头。
4) 取适量混合后的底物液加到膜上。
5) 根据发光强度的强弱调整曝光时间。
2. ELISA检测
1) 根据需要吸取适量底物A液和底物B液1:1等量混合。
注:该试剂现配现用,A液和B液在吸取过程中必须要更换吸头。
2) 微孔板中每孔可加50~200 μL。
3) 10~60 min内检测,40~50 min内检测更佳。
保存条件
2~8℃避光保存,保质期12个月。
注意事项
1. 曝光背景高,可能是膜没洗干净、二抗或一抗浓度过高、封闭液不合适导致,请做适当调整。
2. A液和B液在吸取过程中必须要更换吸头,A液和B液相互污染后会导致其逐渐失效。
3. 如无信号,可能是目的蛋白表达弱,可适当延长曝光时间。
4. 荧光迅速淬灭,可能是目的条带荧光信号过强,导致HRP快速消耗ECL底物,建议降低二抗浓度。
5. NaN3或金属螯合剂会抑制 HRP 活性,溶液中应避免使用。
6. 工作液需现配现用。
